How should I design the constant flanks that Twist will use for PCR amplification?
Please consider the standard rules for primer design:
- Length of 20-25 bp
- 35-70% G/C content
- Melting temperature (Tm) of 55-60°C
- Primer pairs should have a Tm within 5°C of each other
- Primer pairs should not have complementary regions
- Avoid internal primer binding sites in the variable region
