Preparing DNA sequencing libraries often involves complex workflows and only produces small batches of samples at a time. For genomic studies where hundreds of specimens are being evaluated, cost-effective tools that can be integrated with existing processes are essential.
The Twist 96-Plex Library Preparation kit enables high-throughput library construction for whole genome sequencing. The kit is purpose-built for use in high throughput applications, including industrial seed selection, cattle breeding, and rational drug design. With primer tunability for varying GC content and up to 960 samples per kit, the 96-Plex Library Preparation kit offers an optimal balance between scalability and robust library construction.
Workflow
The Twist 96-Plex Library Preparation workflow starts with up to 96 individual samples in a plate. After the first random priming and barcoding step, all samples are pooled into a single tube for the rest of the process. An optional plate barcode can be added in the index read position to allow simultaneous sequencing of multiple sample plates.
Recommended Application
- Low-pass sequencing
- Industrial seed selection
- Cattle breeding
- Plasmid sequence verification
Kit Overview
The Twist 96-Plex Library Preparation kit contains all reagents needed to prepare double stranded libraries with full length adapters compatible with Illumina sequencing chemistries from 50 nanograms of high molecular weight input material. No additional primers, enzymes, or beads are required.
The kit is available in 960 sample format, containing sufficient materials for five or ten 96-sample pooled batches.
At just $10 per sample, the Twist 96-Plex Library Preparation kit scales up without breaking the bank.
Each preparation process produces 96 sequencer-ready libraries that seamlessly integrate with existing
sequencing workflows.
“To be able to pool all of the samples to with 5 tubes instead of almost 500 different wells was really nice!”

GC content is variable with different organisms, which can cause varying biases in library preparation. The Twist 96-Plex Library Preparation kit supplies two primer sets, for high and low GC genomes. By using the primer set suitable for the organism being tested, users can tune the kit to fit their needs.
Sequencing libraries prepared with the Twist 96-Plex Library Preparation kit display uniform read counts across each plate. Bead-based size selection evenly captures fragments within a specified range.
