After cloning, how many colonies should I pick to increase my chances of finding a clone with my correct sequence?

For standard complexity sequences and considering our error rate of 1 in 7500, selecting the recommended number of colonies, as shown in the graph, provides a 95% probability of finding a colony with the correct sequence.

Please note that this recommendation is a starting point. Various factors, such as vector type, sequence complexity, and sequence content, can affect colony growth. In some scenarios, selecting 2 to 3 times the recommended number of colonies may be necessary.
